| 1. | Combine 20ul GeneReleaser® and 1.5ul of heparinized whole blood. |
| 2. | To this add 20ul of GR diluent (made of 15 mls of .01MTris + .05MKCl pH 8.3 + 0.5mls 1% Triton X-100). |
| 3. | Vortex vigerously, overlay with mineral oil and microwave at 4500 Watt-Minutes. A beaker of 150 mls of ambient temperature water may be added to the microwave as a heat sink to prevent the tubes from poping open. |
| 4. | Following microwaving, centrifuge for 30 seconds to pellet GR and debris. Use 1.5ul of supernatant for PCR. In some cases several different sized aliquots of the treated supernatant should be evaluated to determine the optimum quantity to be emplyed to achieve the desired sensitivity. |
