GeneReleaser® DNA AMPLIFICATION FROM DNA AMPLIFICATION FROM CEREBROSPINAL FLUID Protocol

The following suggestions are made as guidelines not knowing the exact details of your circumstances. They may require modification to accommodate your application. Spiking noninfected CS fluids with a model organism may be required to establish suitable protocols. The protocol below works for microbial organisms and intracellular microbial organisms. If an extracellular virus is being examined then the standard GR protocol should be followed using the CSF directly as the specimen material.

| 1. | In a standard amplification tube (0.5ml) dilute 100µl-500µl of CSF. |
| 2. | Centrifuge for 5 minutes to pellet cells and bacteria. |
| 3. | Discard the supernatant. |
| 4. | Wash the pellet with 100µl of 1X PCR buffer by vortexing, centrifuging 1 minute and discarding the supernatant. |
| 5. | Add 20µl of GeneReleaser® to the pellet (usually 10µl pellet volume). |
| 6. | Vortex vigorously to resuspend the pellet and to mix with the GR, overlay with oil and place tubes in microwave rack. |
| 7. | Perform microwave protocol per instructions. |
| 8. | Add appropriate amplification reagents and begin amplification cycles. |
