MTB SAMPLE PREPARATION
|
|
A. FOR NaOH DECONTAMINATED PREPARATION
|
| 1. | Concentrate or enrich MTB by standard method (ASM or other) from
sputum, urine etc. Transfer suitable aliquot to 0.5 ml PCR tube. |
| 2. | If NaOH decontamination method was employed, neutralize excess
NaOH by three washes with 20 volumes (20 x the estimated volume of
the pellet obtained from enrichment) 1M tris buffer pH 8.3. Centrifuge
after each was to repellet the MTB containing material. |
| 3. | Completely remove the supernatant from the last wash. |
| 4. | Treat the remaining pellet with GeneReleaser® according to the microwave procedure and perform PCR. |

|
B. FOR ENRICHED BUT NONDECONTAMINATED SAMPLE MATERIAL
|
| 1. | Transfer suitable aliquot of enriched MTB sample prior to NaOH decontamination step to 0.5 ml PCR tube. |
| 2. | Treat residue with GeneReleaser® according to microwave protocol and perform PCR. |
|
|
Using either of the above methods we can detect 10 MTB organisms.
