GeneReleaser® MTB SAMPLE PREPARATION Protocol
MTB SAMPLE PREPARATION
A. FOR NaOH DECONTAMINATED PREPARATION
1. Concentrate or enrich MTB by standard method (ASM or other) from sputum, urine etc. Transfer suitable aliquot to 0.5 ml PCR tube.
2. If NaOH decontamination method was employed, neutralize excess NaOH by three washes with 20 volumes (20 x the estimated volume of the pellet obtained from enrichment) 1M tris buffer pH 8.3. Centrifuge after each was to repellet the MTB containing material.
3. Completely remove the supernatant from the last wash.
4. Treat the remaining pellet with GeneReleaser® according to the microwave procedure and perform PCR.
B. FOR ENRICHED BUT NONDECONTAMINATED SAMPLE MATERIAL
1. Transfer suitable aliquot of enriched MTB sample prior to NaOH decontamination step to 0.5 ml PCR tube.
2. Treat residue with GeneReleaser® according to microwave protocol and perform PCR.
Using either of the above methods we can detect 10 MTB organisms.
