GeneReleaser® DNA AMPLIFICATION FROM SYNOVIAL FLUID Protocol
Thank you for your interest in GeneReleaser® and its use on synovial fluid. The following suggestions are made as guidelines not knowing the exact details of your circumstances. They may require modification to accommodate your application. Spiking noninfected synovial fluids with a model organism may be required to establish suitable protocols.
1. In a standard amplification tube (0.5ml) dilute 100µl of synovial fluid (or smaller volume) with 200µl of preservative free PBS.
2. Centrifuge for 5 minutes to pellet cells and bacteria.
3. Discard the supernatant.
4. Wash the pellet with 100µl of 1X PCR buffer by vortexing, centrifuging 1 minute and discarding the supernatant.
5. Add 20µl of GeneReleaser® to the pellet (usually 10µl pellet volume).
6. Vortex, overlay with oil and place tubes in microwave rack.
7. Perform microwave protocol per instructions.
8. Add 70µl of a 1.43X master mix to each tube following the conditions for amplification.
